Hydrogen Peroxide Test

The Hydrogen Peroxide is a 72 hour test which is used to indicate the percentage of barley corns that will germinate under near optimal conditions i.e. the number of viable corns within the sample.

Method
  • Place 200 corns in a glass jar
  • Cover with 100ml of 0.75% hydrogen peroxide solution
  • Place in the incubator at 20⁰C for 48 hours
  • Drain after 48 hours and fill with fresh hydrogen peroxide solution
  • Place in the incubator for another 24 hours
  • Count the number of corms that have germinated
  • If the result is less than 98%, place non germinated corns in a petri dish with 4ml of fresh water
  • Incubate for 24 hours and re-check
